Studies on the Expectorant Activities of Water Extract of Semen Benincasae and its Fractions in the Rat Trachea

Abstract
Objective : This study was carried out to evaluate effect of expectorant by semen Benincase an its various fraction in the trachea of rats(SP-D, M. 105g).
Methods : After a single intraperitoneal injection of water extract, 100, 200, 400mg/kg, b.w, respectively, the contents of histamine, acetycholine and serotonin, which are neurotransmitters, exhibited maximum levels at 120 min as a time course in the rat trachea. However, there were no differences of neurotransmitter contents related with the dose of 200 or 400mg. When rats were administrated with the water extract (200mg/kg, b.w), the protein-bound hexose, a major component of mucin or mucus, was secreated about 118% as compared with the control at 120 min in the rat trachea. The period of treatment was scheduled so as to obtain the maximum activity of lysozyme as an important enzyme for the liquefaction of mucus as indicated in time course study.
Results : the results from this approach were in the complete agreement with pattern of hlycoprotein secretion. Microscopic observation after treatment with the water extract or its various fractions (Fr-Ep, A, B) to the tracheal tissue of rats showed that Fr-B possessed a significant effect on the activity of ciliary movement. Administration of the water extract or Fr-B stimulated the secretion of phenol red dye or fluorescein sodium associated with mucus content of tracheobronchial lumen of rats. Especially, the secretion of fluorescein sodium by treating Fr-B had a makedly stimulatory effect (131%) as compared to the control after 120min.
Conclusion : On the basis of this study, it was sugested that Semen Benincasae might be very useful for the stimulation of expectorant activity.
